I think Eastwood probably holds the record for famous quotes.
Eastwood was great, but we probably remember Eastwood better because he's more recent. But IMHO Mae West was the all time best. The actual quote you referenced (she was speaking to a policeman) "Is that a gun in your pocket or are you just happy to see me?"

A few more:

One of my personal favorites, from Peter Lorre in The Maltese Falcon. Lorre, playing the little tough guy, has a gun pointed at Bogart. Bogart manages to punch Lorre in the face (knocking him out) and takes his gun.

Next scene, Lorre comes to, opens his eyes to see Bogart pointing the gun between his eyes from close range. Lorre looks around assessing his situation, then looks down at himself.

"You got blood on my shirt."

Best movie quote ever, in fact the best piece of film ever used. Was Quint's soliloquy in the movie Jaws, about the loss of the Indianapolis, and what the crew went through before being rescued. Gives me chills everytime. Every word is perfect.
